Properties: | This host strain is a derivative of the Raptor strain and can be used for bioengineering where purified products with reduced endotoxicity are desirable. Enterobacterial common antigen is no longer produced and a UDP N-acetylglucose epimerase has been chromosomally integrated increasing the availability of UDP-GalNAc. The genes wecA-wzzE in the ECA synthesis locus were replaced with gne, UDP N-acetylglucose epimerase from the Campylobacter jejuni strain 81116, which was inserted under control of the native, constitutive wecA promoter, in the Raptor background using CRISPR technology. wzzE encodes the enterobacterial common antigen chain length regulator which was removed to prevent any interference with recombinant polysaccharide production. |
